Inflammatory circuit that triggers breast cancer uncovered

ScienceDaily (Feb. 24, 2012) ? Although it's widely accepted that inflammation is a critical underlying factor in a range of diseases, including the progression of cancer, little is known about its role when normal cells become tumor cells. Now, scientists from the Florida campus of The Scripps Research Institute have shed new light on exactly how the activation of a pair of inflammatory signaling pathways leads to the transformation of normal breast cells to cancer cells.

The study, led by Jun-Li Luo, an assistant professor at Scripps Florida, was published online before print by the journal Molecular Cell on February 23, 2012. The scientists' discovery points to the activation of a self-sustaining signaling circuit that inhibits a specific RNA, a well-known tumor suppressor that helps limit the spread of cancer (metastasis). Therapies that disable this circuit and halt this miRNA repression could have the potential to treat cancer.

The Spark that Ignites Trouble

In the new study, scientists identified the specific pathways that transform breast epithelial cells into active cancer cells. The researchers found immune/inflammatory cells ignite the transient activation of MEK/ERK and IKK/NF-kB pathways; the MEK/ERK pathway then directs a consistent activation of a signaling circuit in transformed cells. This consistent signaling circuit maintains the malignant state of the tumor cells. Luo compares this process to starting a car -- a car battery starts the engine much like the transient signal activation turns on the consistent signal circuit. Once the engine is started, it no longer needs the battery.

The scientists go on to show that the initial activation of these pathways also activates IL6, a cytokine involved in a number of inflammatory and autoimmune diseases, including cancer. IL6 acts as a tumor initiator, sparking the self-sustaining circuit in normal breast cells necessary for the initiation and maintenance of their transformed malignant state.

In establishing that self-sustaining signal circuit, IL6 represses the action of microRNA-200c, which is responsible for holding down inflammation and cell transformation. Since enhanced microRNA-200c expression impairs the growth of existing cancer cells and increases their sensitivity to anti-tumor drugs, compounds that disable microRNA-200c repression have the potential to act as a broad-spectrum therapeutic.

Interestingly, the new findings dovetail with the "multiple-hits theory" of tumor formation, which posits that once normal cells in the human body accumulate enough pre-cancerous mutations, they are at high-risk for transformation into tumor cells. While the newly described initial pathway activation is momentary and not enough to cause any lasting changes in cell behavior, it may be just enough to tip the cell's transformation to cancer, especially if it comes on top of an accumulation of other cellular changes.

WASHINGTON (AP) ? The reviews are in, and Mick Jagger sizes up President Barack Obama's singing of the blues this week as nothing short of "courageous."

The Rolling Stones frontman said Thursday the president did "very well" when bluesman Buddy Guy successfully prodded him to join in on a few lines of "Sweet Home Chicago," the blues anthem of the president's home town, during an East Room concert earlier in the week.

"I thought he was courageous, because there was some element of reluctance," Jagger said, adding that "it was a hard moment for the president to get out of."

Jagger, in a phone interview with the AP, also said he was pleased to see that first lady Michelle Obama was wearing a dress that night designed by L'Wren Scott, his longtime girlfriend, who had a front-row seat for the concert.

Jagger, 68, said he's never been invited to play at the White House before ? or for any head of state ? and he rated it a "surreal gig experience."

"I loved it," he said. "It was really unusual, surreal, and strange, but very enjoyable. ... I went out there and I tried to put out my best."

What's next for Jagger? He said he's on a minivacation on the East Coast with his kids, and they were heading out the door to play soccer.

But will the Stones get together this year for a 50th anniversary tour?

"I hope we certainly do something," he said. "It would be very appropriate, I think, and I look forward to doing something. But nothing is yet booked."

Jagger and other blues greats played the East Room on Tuesday as part of an "In Performance at the White House" concert that will air Monday on PBS.

LONDON (AP) ? The closing ceremony for 2012 London Olympics will be themed "A Symphony of British Music" ? with organizers promising a hit-packed journey from classical composer Edward Elgar to Grammy soul diva Adele.

"It is, in a sense, the soundtrack of our lives," said Stephen Daldry, executive producer of the Olympic and Paralympic opening and closing ceremonies.

The ceremony's artistic director, Kim Gavin, said the Aug. 12 show would be an "elegant mashup of British music" packed with recognizable songs arranged symphonically. The London Symphony Orchestra will provide musical backing.

"We go from ? without saying we've booked anyone ? Elgar to Adele," said music director David Arnold.

The creative team is keeping the names of the musicians involved under wraps, although The Rolling Stones, Paul McCartney and the Spice Girls have all been suggested as possible performers.

Olympic officials did not say whether Adele, Britain's hottest musical export of the moment, had been asked to take part. The singer-songwriter's album "21" has spent a full year on the charts, winning six Grammys earlier this month, including album of the year. Adele also won two trophies Tuesday at the Brit music awards.

Gavin, who has overseen tours for the band Take That and directed the 2007 Princess Diana memorial concert, said the London Olympics closing ceremony would be "the biggest after-show party ? the show being the sport."

Once we decided to go with a brown countertop for the vanity, we left behind the black and white color scheme we had decided on and went in a different direction. ?

I have a confession to make. I love self-improvement books. Back in my yuppie days, I spent hours pouring over books on time-management, listening skills, presentation skills and more. Out of the hundreds of business books I read while working for the electric cooperative program, the one that made the greatest impact on me was Stephen Covey's The Seven Habits of Effective People. Twenty years after I first read it, the book still sits on my favorite's bookshelf. I still use a weekly calendar based on his approach, and I still strongly believe in having a personal mission statement.

But the mission statement of a young woman focused on family, career and community is very different from that of a middle-aged woman exploring her long-dormant artistic side. It took awhile, but I finally feel confident that I know where I'm heading. Here's my new mission:

Egypt inquiry blames fans, police for stadium deaths (Reuters)

CAIRO (Reuters) ? An Egyptian parliamentary inquiry into the deaths of 74 people in a soccer stadium disaster has found both fans and lax security to blame for the worst incident of its kind in the country's history, the legislator leading it said on Sunday.

The preliminary findings of the inquiry into the Port Said deaths offered little to support the view of those Egyptians who believed the deaths were the result of a plot hatched by elements within the establishment seeking to create chaos.

Ashraf Thabet, the member of parliament who headed the inquiry, listed factors including incitement by sports TV channels as the cause of the incident which touched off several days of violent protests in which 16 more people were killed.

The incident occurred at the end of a match between Port Said-based al-Masry and Cairo's Al Ahly, the most successful club in Africa.

Witnesses to the February 2 incident had said hundreds of al-Masry supporters surged across the pitch to the visitors' end causing panicked Ahly fans to dash for the exit. But the steel doors were bolted shut and dozens were crushed to death in the stampede. Many believe the incident was sparked by hired thugs.

Thabet spoke of instigators who had used thugs and hardcore soccer fans to take "advantage of the tension surrounding the game to achieve some political gains," but without giving further details. "We will announce their names," he said.

Critics of the military-led authorities had laid the blame on the government, some saying the violence was planned to create an air of chaos that would add to the case for army rule.

Thabet said fans were not inspected while entering the stands and there was a lack of order inside and outside the stadium. "Security facilitated, allowed and enabled this massacre," he said. The security forces had failed to predict trouble and control the crowd stampede, he added.

He also laid blame on "Ultras," the hardcore soccer fans who regularly confront riot police at matches and have been on the front line of confrontations with the security forces since the uprising that toppled Hosni Mubarak from power.

Most Egyptian teams have their own group of Ultras.

Thabet said there had been tension between the fans before the match. "Both ultras and thugs attacked Ahly fans and this is part of Ultras' culture," Thabet said.

Similar instances of pitch invasions had occurred in Port Said in the past few months, Thabet said, but without causing casualties.

Demonstrators blaming the security forces for the deaths targeted the Interior Ministry in downtown Cairo in protest at what some saw as police complacency. Others said the security forces had a role in igniting trouble.

The committee said investigations were still ongoing and that it would announce final results in its final report which would assign political responsibility for the events.
